Yes, the symbol ® can only be used after your trademark is successfully registered with an IP office.
Please keep in mind that you are allowed to use the ® only with the exact same version of your trademark you registered with the respective IP office. In case you use an iteration or a part of the trademark, you can use the symbol ™ instead. If your trademark is pending, you can also use the symbol ™ to indicate that you are in the process of obtaining a trademark registration.
Also, you can only use the ® symbol in countries where you hold your registration.
