No, nationwide commercial activity is not required to register a trademark. In fact, in most jurisdictions, only a bona fide intent to use the mark in commerce is sufficient.

The only jurisdiction where market activity must be demonstrated is the US, and even there, activity in a single state is sufficient.

Trademarks also apply to brick-and-mortar stores. Even if sales are conducted locally, a registered trademark is valid nationwide in the jurisdiction where it is registered.
